I would have had the scene where Adam Sandler's character dies (that always makes me cry) and I'd have that as Number 1.
Number 2 would definitely go to Planes, Trains and Automobiles. It's such an underrated movie and I love it (it's my dad's favourite movie, too).
Mrs. Doubtfire is definitely Number 3.

Looking through all of the other comments, I think, just maybe you should make a part 2. So many missed, i.e. Paul Blart, Mall Cop as he's browsing the dating site. Real lonely guy moment there.

Ah man, Click always gets me. Especially that scene with his father and the scene where he's dying. It's so depressing, thinking about how eventually you'll die and there's nothing you can do about it.
